ABSTRACT
Investigation on cell line T47D using extract Pandanus conoideus red variety
showed the extract could reactivate p53 mutation. This effect was expected caused
by interaction of -tocopherol dan -carotene which it’s contain (Astirin and
Budiani, 2007). Pandanus conoideus yellow variety has known containing
tocopherol higher than red variety (Budi and Paimin, 2005). Other investigation
showed LC50 P.conoideus yellow variety extract was at concentration 0,25µl/ml
(Pratiwi, unpublished). Wibowo et al (2007), showed that lauric acid from VCO
has cytotoxic activity on cell line T47D, MCF7, HT29, Raji, and HeLa. The aim of
this investigation has examined potentiation effect of cytotoxicity and
antiproliferative activity of combination of P.conoideus yellow variety extract and
lauric acid from VCO, and found effective concentration in proliferative inhibition
on breast cancer cell line T47D
This investigation included cytotoxicity and doubling time tested by direct
counting method. In cytotoxicity test, the cells had incubated for 24hr in RPMI
1640 media after added combination of yellow variety extract and lauric acid from
VCO in six group i. e control group and six concentration of lauric acid (0,1µl/ml,
0,05µl/ml, 0,025µl/ml, 0,0125µl/ml and 0,00625µl/ml) combined 0,25µl/ml
P.conoideus. Proliferatif inhibition activity has known by doubling time test for
72hr. Cell amount counted at 0, 24, 48, and 72hr.
Investigation resulted LC50 of combination of P.conoideus yellow variety
extract and lauric acid from VCO was in 0,25µl/ml P.conoideus and 0,0125µl/ml
lauric acid. Effective concentration on doubling time test was found in 0,125µl/ml
P.conoideus and 0,00625µl/ml lauric acid. The investigation also showed hat the
combination has sinergistic effect in cytotoxic and inhibition than using
P.conoideus yellow variety extract and lauric acid alone.
