UJI ANTIBAKTERI KOMPONEN BIOAKTIF DAUN LOBAK (Raphanus sativus L.) TERHADAP Escherichia coli DAN PROFIL KANDUNGAN KIMIANYA

ABSTRACT
Indonesia has many kind of plants, which some of them are medicinal
plants, and it have been used to cure various disease. One of medicinal plants is
Radish (Raphanus sativus L.), known as vegetable. That’s the reason conducting
some research to find the bioactive compound from plants, which can used as raw
material of drug. Another reasons are costly drug prices in market, the death case
due to microbial infections, and the increasing of bacterial resistance because of
uneffectiveness usage of antibiotics. This study was aimed to know the
antibacterial effect of bioactive compounds from Radish’s leaves against
Escherichia coli.
Bioactive compounds were found by extraction dan partition using
sentrifugation. Dry powder from Lobak leaves was extracted by chloroform and
methanol. After the antibacterial tested, methanol extract given an antibacterial
effect so it had separated in partition by ethyl acetate into the soluble and
insoluble part. The soluble part of ethyl acetate given an antibacterial effect on
concentration 30-50%, shown by clear zone (average 10 mm). It had separated
again into soluble and insoluble fraction with dicloromethane. Dicloromethane’s
soluble fraction formed clear zone on 10% concentration (8,30 mm) and 8,42 mm
on 20%. Then the chemical constituen’s profile of the most active fraction has
been monitored using KLT method and detected with spray chemicals.
The result indicating that Radish’s leaves have bioactive compound from
fenolic group. It has been proved by ammonia detection which was gave a yellow
colored, and dark blue with FeCl3 detection.
