ABSTRACT
In Indonesia, breast cancer is the second highest killer after cervical cancer. Most
likely, in breast cancer case also stimulate from gene mutation of p53. Genetic changes of
p53 resulting of programmed death cell (apoptosis) lost, cause of cell display
uncontrolled growth. Many cancer treatment like surgery, chemotherapy, and
radiotherapy was uneffective. That is caused many alternative treatment such as herbal
medicine.Virgin Coconut Oil (VCO) known that it has some lauric acid which neutralize
free radical, it hopes that lauric acid can be use as cancer resistant madicine. The
objective of this research is to find out the in vitro effect of the lauric acid on the
antiproliferation, apoptosis and expression of p53 of breast cancer T47D cell lines in
RPMI 1640 medium.
Direct counting to measure the growth inhibitory effect, examined by staining the
viable treated cells using trypan blue. The proliferation kinetic was determined by
counting the cell number in time courses to determine the doubling time of th e cells.
Apoptotic proscess was examined by observaring microscopally the cell morphology that
was stained using immunocytochemistry with primary monoclonal antibody anti p53 and
secondary antibody anti sheep IgG biotin conjugate
As the result, lauric acid are able to decrease cell proliferation, which is 1,09
bigger on 0,003125μl/mL concentrate, 1,27 bigger on 0,00625μl/mL and 1,75 bigger on
0,0125μl/mL concentrate, comparison with control. Luric acid able to decrease p53
expression decline mitotic and induced apoptosis
