PERTUMBUHAN DAN KANDUNGAN AJMALISIN PADA KULTUR KALUS DAUN TAPAK DARA (Catharanthus roseus (L.) G. Don.) DENGAN PEMBERIAN KINETIN DAN CEKAMAN ASAM

ABSTRACT
The aim of this research is to study the effect of variation of kinetic and
acid stress on growth and ajmalicine content on leaf callus culture Catharanthus
roseus (L.) G. Don.
The research methods used in this research is factorial experiment 4x4.
The first factor is HCl treatment at 4 level of acidity, they are pH 2,9; 3,9; 4,9;
and 5,9 as a control. The second factor is variation of kinetic consentration each 0
mg/L as a control; 50 mg/L; 100 mg/L; and 150 mg/L. Each treatment (pH and
kinetic combinations) consist of 3 levels. The collected data consist of qualitative
data (callus morphology) presented descriptively and quantitative data (fresh
weight callus, growht rapid callus, and dry weight callus). The ajmalicine
compound tested qualitatively with thin layer chromatography (TLC), then
followed the quantitatively test with thin layer chromatography-scanner. Analysis
the quantitative data with Analysis of Varian (Anava) then Tamhane research
level 5%.
The result of research showed that variation of kinetic consentration
influence fresh weight callus and growth rapid callus, but didn’t influence dry
weight callus. Variation of pH didn’t influence fresh weight callus, growht rapid
callus, and dry weight callus. The highest ajmalicine content in callus culture
Catharanthus roseus (L.) G. Don., didn’t syncronize with the highest variation of
pH consentration and kinetic treatment. The highest ajmalicine content product
were in the Murashige Skoog (MS) medium with influence pH 5,9 and kinetic
consentration 100 mg/L.
