UJI TOKSISITAS HASIL FRAKSINASI DAUN RUMPUT MUTIARA (Hedyotis corymbosa (L.) Lamk.) TERHADAP Artemia salina Leach DAN PROFIL KANDUNGAN KIMIA FRAKSI TERAKTIF

ABSTRACT
Cancer is still become one of the most daunted disease by society at
present. But until now it still haven’t been found the way to overcome the disease
by satisfied. The other way selected by some patient of this disease by exploiting
natural product. One of the plant which have potency for the commodity of
traditional drug is Rumput Mutiara ( Hedyotis corymbosa ( L.) Lamk.). In
Indonesia this plant has not been used optimally yet as one of the healing form,
although the plant has many benefit for healing of various disease, including to
cure breast cancer ( Hambali, 2005). The use of this traditional drug requires to be
proved scientifically.
In this research the toxicity test from fractionation result of Rumput
Mutiara (Hedyotis corymbosa ( L.) Lamk.) leaves was conducted by using Brine
Shrimp Lethality Test ( BST) method to know its potency as drug of anticancer
and compound fraction content on the most active profile. Fractionation method
that has been used by Vacuum Liquid Chromatography (VLC). The test has been
done by including ten larva A. salina Leach into flacon contain test sample. The
mortality percentage of A. salina Leach larva was counted 24 hours after giving of
test sample rate series then made equation of linear regression to determine values
of LC50-24 hours.
The result of toxicity test showed that chloroform extract more toxic than
methanol extract. The partition result of chloroform extract showed that soluble
fraction of ethyl acetate more toxic than insoluble fraction of ethyl acetate.
Dissolve fraction of ethyl acetate was used as toxic fraction, then be fractionated.
The test result of each fraction by BST showed that fraction VI is the most toxic
with values of LC50-24 hours = 281,77 µg / ml. TLC analysis by various specific
reagent showed that compound of alkaloid, flavonoid, terpenoid and ursolic acid
was found in the fraction VI.
