PERTUMBUHAN TUNAS PURWACENG (Pimpinella alpina K. D. S.) PADA PERLAKUAN PRAKONDISI DENGAN VARIASI KONSENTRASI THIDIAZURON

ABSTRACT Purwaceng (Pimpinella alpina K. D. S.) is one of endangered species which is potential to be developed as aphrodisiac source. To support conservation and cultivation of purwaceng, it is needed to prepare the plant material. Tissue culture technique is one elternative methode for both cultivation and regeneration. Constraint in micropropagation of purwaceng are the limited explant source, high level of senescence and low level of shoot multiplication The aims of this research were to increase shoot growth by in vitro preconditioning on Driver and Kuniyuki Walnut (DKW) medium with various consentration of Thidiazuron (TDZ) (0 mg/l; 0.5 mg/l; 1 mg/l; 1.5 mg/l; 2 mg/l). The cultures were incubated in preconditioning medium for 2 weeks, then they were subcultured on DKW basal medium for 8 weeks. Wet and dry weight, lenght and also visual performance of shoot were observed. During incubation in preconditioning medium, there was no senescence on shoot. Observation after 10 weeks incubation, showed that the treatment has no significant effect on shoot growth, but succes to delayed senescence. Wet weight, dry weight and length of shoot at preconditioning medium were generally lower than control. Precondition with TDZ at consentration 0 mg/l to 1.5 mg/l increased biomass production of purwaceng culture wich indicated by increasing wet and dry weight, and decreasing at consentration 2 mg/l of TDZ. The length of shoot increased at consentration 0 mg/l to 1 mg/l, but it decreased at consentration 1.5 mg/l and increased again at consentration 2 mg/l of TDZ. The highest wet and dry weight was showed by treatmen at 1.5 mg/l TDZ, but lowest on lenght of shoot. Keywords: Pimpinella alpina K. D. S., in vitro, preconditioning, thidiazuron, shoot growth.
