ABSTRACT The aim of the study was to know the proline and polyphenol content of tannia (X. sagittifolium) in various light intensity and water availability treatments. Proline and polyphenol are some of secondary metabolites which have a role in plant defence mechanism under stress. Proline act as osmoprotectant whereas polyphenols are antioxidant compound which act as radical scavenger. The research done in randomize complete block design with two factors and three replications. The first factor was light intensity in three levels (N0= 100%, N1= 45% and N2= 25%), the other factor was water availability in four different field capacities (A1= 40%, A2= 60%, A3= 80% and A4= 100%). The treatments done by watering the plants once in two days under the paranet condition for 45 days. There were two parameters measured (proline and polyphenol). The proline content measured by ninhydrine method whereas the polyphenol by folin cioucalteu. The datas obtain were analyzed by General Linear Model Univariate followed by DMRT in 5% confident level. The results showed that the proline content significantly affected by the combinations of light intensity and water availability, but the polyphenols didn’t. The highest proline content shown by N0A3 plant at the average level of 4,1388 µmol/g FW, and the lowest shown by N0A2 plant at the average level of 1,9496 µmol/g FW. The highest polyphenol content shown by N0A4 plant at the average level of 0,7029 mg of gallic acid/g DW, and the lowest shown by N2A4 plant at the average level of 0,3993 mg of gallic acid/g DW.
