VARIASI GENETIK UBI JALAR (Ipomoea batatas Linn.) MAGELANG BERDASARKAN POLA PITA ISOZIM

ABSTRACT
Sweet potato (Ipomoea batatas) is one of the important ingredients
commodity in Indonesia. The local varieties of sweet potato in Magelang such as
Sembowo, Jegros and Merketek have a big potential but until now it was never
research on molecular stage. The research aim was to study the genetic diversity
and genetic distance from the peroxydase and esterase isozymes analyse with
electrophoresis method.
The research was conducted in Biotechnology Forest Laboratory, Forestry
Faculty of Gajah Mada University on January until March 2008. There are 9
samples bud of leaves was put from each varieties. The samples was extracted
with extract buffer and then samples was centrifuged with 1500 rpm velocity until
20 minutes. Next 10 µl supernatan was put and take on to the electrophoresis gel.
Poliacrylamides gel was used are running gel and separated gel. Next the samples
was electrophoresed with 85 voltage until 3 hours and 100 mA then gel was
moved on to the staining plate for colouring with peroxydase and esterase
enzymes. After that was conducted fixation process until 24 hours at 4 °C and
then gel was dried and looked on.The genetic diversity was knowable from the analyse result of isozyme
banding pattern and calculated the genetic diversity of parameters. And then
genetic distance was knowable based on the matric of genetic distance which
applied on the shape of dendogram with UPGMA (Unweighted Pair Group
Method With Arithmatic Averages) method and calculated with SPSS 13 software.
The result of the research showed that genetic diversity of sweet potato
Sembowo, Jegros and Merketek varieties. It was showed from polymorphic locus
was produced, they are POD-1 loci and EST loci. Based on clustering samples
was produced the same dendogram not only peroxydase but also esterase
dendogram which showed that Jegros and Merketek varieties have nearest the
genetic distance that on resemblance number 0 and so far the genetic distance
with Sembowo varieties that on resemblance number 25.
