PENGARUH PEMBERIAN EKSTRAK BUAH MERAH (Pandanus conoideus Lam. ) TERHADAP TINGKAT FERTILISASI DAN PERKEMBANGAN EMBRIO TAHAP PREIMPLANTASI MENCIT (Mus musculus) IN VIVO

Red fruit extract (RFE) with its alpha tocopherol and betacaroten functioning as antioxidant are expected able to prevent the free radical on the female reproducing system. Due to the limited information concerning on the influence of RFE to the reproduction function specifically at the growth of embryo, a study is then required in order to know the influence of RFE to the level of fertilization and growth of mice embryos during the preimplantation phase. The application of RFE dose 0mL, 0,05mL, and 0,1mL for seven days is done to know its influence to the level of fertilization, then the application of RFE is continued during 0-3 days stage of pregnancy (0 day of pregnancy is the formation of the vaginal plug). It is done to know the influence of giving RFE to the level of fertilization and growth of mice embryos during the pre-implantation phase. The super-ovulation of the white mice is done by inseminating 5 IU PMSG and hCG (48 hours after PMSG), breeding those mice with male and female comparison = 1:1. The single-cell-embryos are collected 20 hours after the inoculation of hCG by incising of the oviduct at the CZB+Hepes+hyaluronidase medium, then, the egg cells are cleaned in same medium without hyaluronidase. The ovum is fixated by using glutaraldehyde 2,5% in PBS pH 7,4, soaked in neutral formalin 10%, dehydrated by using ethanol of 95%, coloured with lacmoid 0,25% in acetic acid of 45%, the ovum are then cleaned by applying acetoglycerol and is done by observing the pro-nucleus. The morula and blastosis are collected after 84 hours after the inoculation of hCG, each uterus is then flushed by applying CZB+Hepes medium, embryos are cleaned by the same medium, and the last is the observation towards those embryos. The influence of the red fruit extract (RFE) at level of white mice fertilization, with dose 0,1mL significantly gives a positive effect (46,01%), when it is compared with dose 0,05mL (37,51%) and controlled (16,38%) towards the formation of the male pro-nucleus. The influence of RFE towards the formation of the female pronucleus at dose 0,1mL (50,91%) and 0,05mL (62,49%), in fact, significantly gives a negative effect (83,62%) at the control (54,16%). The influence of RFE to the growth of embryo during the preimplantation phase with dose of 0,05 mL and 0,1mL may inhibit the growth of mice embryos during the preimplantation phase.
