Pengaruh pemberian kurkumin terhadap derajat inflamasi usus pada mencit balb/c model sepsis paparan cecal inoculum

ABSTRAK Tujuan : Penelitian ini bertujuan untuk mengetahui pengaruh pemberian kurkumin terhadap derajat inflamasi usus pada mencit Balb/C model sepsis paparan cecal inoculum (c.i.). Metode : Jenis penelitian ini adalah eksperimental laboratorik dengan post test only control groups design. Hewan uji yang digunakan adalah 27 ekor mencit Balb/C jantan yang dibagi dalam tiga kelompok, masing-masing 9 ekor. Kelompok K sebagai kontrol, K1 (Kelompok Sepsis) diberi material c.i. 0,1 ml/mencit/hari/i.p. dan K2 (Kelompok Sepsis+Kurkumin) diberi material c.i. dan kurkumin dosis 0,1 ml/mencit/hari/oral. Pada hari ke-8 semua mencit dikorbankan, diambil ususnya, kemudian dibuat preparat histologisnya dengan menggunakan pewarnaan Hematosiklin Eosin untuk menentukan derajat inflamasinya. Data yang diperoleh dianalisis dengan uji statistik Krusskal-Wallis dan dilanjutkan dengan Mann-Whitney, menggunakan program SPSS for Windows Release 16. Perbedaan dikatakan signifikan bila p<0,05. Hasil : Penelitian menunjukkan derajat inflamasi usus pada kelompok K yaitu grade 0 (44,44%), grade 1 (33,33%), dan grade 2 (22,22%). Kelompok K1 yaitu grade 3 (33,33%), dan grade 4 (66,67%), sedangkan kelompok K2 yaitu grade 2 (33,33%), grade 3 (55,56%) dan grade 4 (11,11%). Terdapat perbedaan yang bermakna antara kelompok K1 dan K2 (p= 0,010), kelompok K dan K1 (p=0,000), dan kelompok K dan K2 (p=0,001). Simpulan : Kurkumin dapat menurunkan derajat inflamasi usus pada mencit Balb/C model sepsis paparan cecal inoculum. ABSTRACT Objective : This experiment was aimed to know the effect of curcumin with inflammation grade of intestinal in Balb/C mice sepsis model cecal inoculation. Methods: This research is experimental laboratoric with post test only control group design. The subject is 27 male Balb/C that were divided three groups of mice with 9 mice each group. K Group as control. K1 Group (Sepsis) was given cecal inoculum material 0,1 ml/mice/day/i.p. and K2 Group (Sepsis+Curcumin) was given cecal inoculum material 0,1 ml/mice/day/i.p. and Curcumin with doses 0,1 ml/mice/day/oral. On day 8, subject were sacrified to take their intestine, were made histology slide by Hematosiklin Eosin staining for determining grading inflammation. Statistical analysis was performed with SPSS for Windows Release 16. The data was analyzed with Krusskal-Wallis methode continued by Mann-Whitney methode to determine significant differences, values of p<0,05 were considered statistically significant. Results: The study showed that K Group were grade 0 (44,44%), grade 1 (33,33%),and grade 2 (22,22%). K1 Group were grade 3 (33,33%), and grade 4 (66,67%). While K2 Group were grade 1 grade 2 (33,33%), grade 3 (55,56%) and grade 4 (11,11%). There was significant difference among K1 group and K2 group (p=0,010), among K group and K1 group (p=0,000), and among K group and K2 group (p=0,001). Conclusion: This study concluded that curcumin can decrease the inflammation grade of intestinal in Balb/C mice sepsis model cecal inoculation.
