Pengaruh pemberian seduhan rosela (hibiscus sabdariffa) terhadap kadar kolesterol total darah tikus putih (rattus norvegicus)

ABSTRAK
Berbagai penelitian di dunia telah membuktikan bahwa ekstrak rosela (Hibiscus sabdariffa) dapat menurunkan kadar kolesterol total darah, tetapi banyak masyarakat mengkonsumsi rosela dengan cara diseduh. Penelitian ini dilakukan untuk mengetahui adanya efek seduhan rosela (Hibiscus sabdariffa) dalam mencegah peningkatan kadar kolesterol total darah tikus putih (Rattus norvegicus).
Penelitian ini bersifat eksperimental laboratorik dengan desain Randomized Controlled Trial (RCT) yaitu pretest and posttest controlled group design, dilakukan di Laboratorium Biokimia Universitas Sebelas Maret Surakarta. Subjek penelitian adalah tikus putih jantan (Rattus norvegicus) sebanyak 30 ekor, strain Wistar, umur 3 bulan, berat badan kurang lebih 200 gram. Tikus-tikus tersebut dibagi menjadi 3 kelompok secara random, masing-masing kelompok terdiri 10 ekor tikus. Semua kelompok diberi pakan tinggi kolesterol. Kelompok I sebagai kontrol, sedangkan kelompok II dan kelompok III diberi seduhan rosela dengan dosis 36mg/200gram BB/hari dan 54mg/200gram BB/hari. Semua tikus diperiksa kadar kolesterol total darahnya setelah masa perlakuan selama 28 hari.
Hasil uji one-way ANOVA menunjukkan tidak adanya perbedaan kadar kolesterol total darah yang signifikan dengan nilai p= 0,126 (p>0,05) pada ketiga kelompok hewan coba. Namun, hasil uji t berpasangan pretest dan posttest tiap kelompok menunjukkan adanya perbedaan kadar kolesterol total darah yang signifikan.
Simpulan penelitian ini adalah tidak ada pengaruh seduhan rosela (Hibiscus sabdariffa) dengan dosis 36mg/200gram BB/hari maupun dosis 54mg/200gram BB/hari dalam mencegah peningkatan kadar kolesterol total darah tikus putih (Rattus norvegicus).

ABSTRACT
Many research in the world has proven that the extract of roselle can decrease the total serum cholesterol level, but most of people consume roselle as steepings. The study was conducted to evaluate the effects of steeped roselle (Hibiscus sabdariffa) in preventing the increasing of total serum cholesterol level of white rats (Rattus norvegicus).
This experimental laboratoric research with randomized controlled trial design, pretest and posttest controlled group design, had been done in Biochemistry Laboratory of Sebelas Maret University, Surakarta, Indonesia. The subjects research are 30 male white rats, Wistar strain, 3 months old, and about 200 gram of weights. Rats were divided into 3 groups, each group consists of 10 rats. All groups were fed high cholesterol food. Group I is as the control group while group II and group III were given steeped roselle 36mg/200 gram body weight/day and 54mg/200 gram body weight/day. Total serum cholesterol level of all rats were tested after 28 days treatment.
The result of one-way ANOVA test revealed that there was no significant difference in total serum cholesterol level between 3 groups, with p= 0,126 (p>0,05). But, the result of t paired test between pre and posttest of each group showed significant difference in total serum cholesterol level of white rats.
In conclusion, steeped roselle (Hibiscus sabdariffa) 36mg/200gram body weight/day dosage and 54mg/200gram body weight/day dosage do not have effects in preventing the increasing of total serum cholesterol level of white rats (Rattus norvegicus).
