Pengaruh pemberian jus stroberi terhadap kerusakan histologis hepatosit mencit akibat pemberian asetaminofen

ABSTRAK Stroberi dengan senyawa fenoliknya memiliki aktivitas antioksidan. Asetaminofen menghasilkan metabolit reaktif NAPQI melalui proses hidroksilasi di hepar yang mengandung radikal oksigen bebas dan bersifat oksidan bagi sel, sedangkan jus stroberi melindungi hepar akibat pemberian asetaminofen. Tujuan penelitian ini untuk mengetahui pengaruh pemberian jus stroberi terhadap kerusakan histologis hepatosit mencit akibat pemberian asetaminofen. Penelitian ini merupakan penelitian eksperimental laboratorik dengan rancangan penelitian posttest only control group design. Hewan uji yang digunakan adalah mencit jantan Swiss Webster berumur  2-3 bulan dan berat badan  20 gr. Hewan uji berjumlah 30 ekor, terbagi dalam 3 kelompok. Kelompok kontrol diberi diet standar selama 14 hari percobaan. Kelompok perlakuan I diberi asetaminofen dosis 3,38 mg/ekor/hari pada hari ke-11 – 14 . Kelompok perlakuan II diberi jus stroberi dosis 624mg/20gramBB/hari selama 14 hari dan asetaminofen dosis 3,38 mg/ekor/hari pada hari ke-11 – 14. Pada hari ke-15 hewan coba dikorbankan, diambil lobus kanan hepar dan dibuat preparat histologi. Gambaran histologis hepar dilihat dengan mikroskop cahaya terang perbesaran 400x. Data yang diperoleh dianalisis menggunakan uji statistik Oneway ANOVA dan dilanjutkan dengan LSD (Least Significant Difference) menggunakan program SPSS 16 dengan α = 0,05. Hasil uji statistik Oneway ANOVA menunjukkan adanya perbedaan pada ketiga kelompok penelitian secara bermakna p:0,000 (p<0,050). Hasil uji LSD menunjukkan adanya perbedaan yang signifikan antara kelompok kontrol-perlakuan I, dan perlakuan I-perlakuan II dengan masing-masing p:0,000 (p<0,050). Simpulan dari penelitian ini adalah pemberian jus stroberi dapat mengurangi kerusakan histologis hepatosit mencit akibat pemberian asetaminofen . ABSTRACT Strawberry phenolics consist of large polymeric compounds have antioxidant abilities. Sub toxic dose of acetaminophen produce reactive metabolic NAPQI through hydroxylation process in liver, containing free reactive oxygen as an oxidant for cells, while strawberry juice protects liver cells from the damage. The object of this research is knowing the effect of strawberry juice to liver cell damage in mice caused by acetaminophen toxicity. This research is a laboratory experimental research with the posttest only control group design. Thirty male mice (Mus musculus) furrow Swiss Webster with age 2-3 months and weight 20 gr which used in this research were divided into three groups. First group was used as control group, second group as treatment group which given acetaminophen dose 3,38 mg/mice/day at 11th - 14th day and third group treatment group II was given strawberry juice dose 624 mg/20 gram BW/day for 14 days and acetaminophen dose 3,38 mg/mice/day at 11th - 14th day, one hour later. At 15th day, mice were killed to take the right liver lobe and make histological specimen. The histological damage of hepatocyte of mice was seen with light microscope using 400x enlargements. All data from measuring in this research were analyzed by Oneway ANOVA statistic test and LSD (Least Significant Difference) test by SPSS program 16th version with α = 0,05. The result of Oneway ANOVA statistic test shows that there is significant difference between three groups p: 0.000 (p<0.050). The results of LSD test shows that there is significant difference between control-treatment group I and treatment group I-treatment group II with p:0.000 (p<0.050) for each group. In conclusion of this research is strawberry juice may reduce liver cells damage in mice which induced by acetaminophen.
