Pengaruh diet singkong (Manihot esculenta) terhadap struktur histologi pankreas tikus putih (Rattus norvegicus)

Abstrak Metode Penelitian: Penelitian ini bersifat analitik eksperimental. Menggunakan rancangan The Post Test Only Control Group Design. Sampel yang digunakan yaitu 27 ekor tikus putih jantan (rattus norvegicus) yang dibagi dalam 3 kelompok: Kontrol, Perlakuan I (PI), Perlakuan II (PII). Kelompok kontrol mendapat diet pakan standar berupa pellet ad libitum. Kelompok perlakuan I (PI) mendapat diet singkong (Manihot esculenta) ad libitum Perlakuan II (PII) mendapat diet pakan campur antara singkong (Manihot esculenta) dan pelet dengan perbandingan 1:1.perlakuan dilakukan selama 6 minggu. Setelah perlakuan semua hewan percobaan dikorbankan dengan cara cervical dislocation kemudian diambil organ pankreas. Untuk selanjutnya dibuat preparat dengan metode block parafin kemudian dilakukan pengecatan dengan Hematoksilin Eosin (HE). Pengamatan preparat dilakukan denganmikroskop. Gambaran pada sel-sel pankreas diklasifikasikan menjadi normal, kerusakan ringan, kerusakan sedang, dan kerusakan berat. Data yang diperoleh dianalisis denngan menggunakan uji statistik Kruskal Wallis yang dilanjutkan dengan Uji Statistik Mann Whitney. Hasil Penelitian: Berdasarkan uji statistik diperoleh hasil terdapat perbedaan bermakna antara kelompok K dengan PI (p=0,001), dan PI dan PII (p=0.002), sedangkan pada K dan PII (p=0. 609) tidak didapat perbedaan bermakna. Simpulan: Pemberian diet singkong dapat mempengaruhi struktur histologis pankreas tikus putih (Rattus norvegicus. Perubahan yang terjadi berupa peningkatan fibrosis. Penambahan asupan makanan lain mempengaruhi peningkatan jaringan fibrotik tersebut. Objective: Cassava is important raw material to make Indonesian traditional food. Cassava (Manihot esculenta) which contains high of carbohydrate and low of lipid and protein level. Continuous cassava consumption without intake of other food can leads to lack of protein condition. Malnutrition Related Diabetes Mellitus (MRDM) ever known as a condition caused by long period of deficit protein intake. Pancreas is organ that related to diabetes mellitus. this experiment aims to find out whether cassava diet can causes histologic structure alteration of white rat’s pancreas. Methode: This experimental analytic research uses The Post Test Only Control Group Design. The research subject were 27 white male rats and devided into 3 groups: control, treatment 1, treatment 2, the conrol group feeded with standard food ad libitum, the treatment 1 group with cassava diet, and treatment 2 with combination of cassava and standard food in 1:1 proportion. This treatment given during 6 weeks. After this treatment all subjects were sacrified by cervical dislocation and the pancreas were taken and made as preparat by block parafin method. and painted with Hematoksilin Eosin (HE). Preparat observation was done with microscope. description of pancreatic cells classified to be normal, light damage, moderate damage, and heavy damage . the collected data analyzed with kruskal wallis test and then mann whitney test. Result: Based on this statistic tests, the result were significant differences between control group and P1 group (p = 0,001), and between PI and PII group(p=0,002), and not significant differences betweencontrol group and PII group(0,609). Conclusion: Cassava diet can influence pancreatic histology structure of white rat’s. the alteration was an increasing of the fibrotic degree. Another food intake can efects the number of fibrotic.
