Pengaruh Sari Buah Kiwi (Actinidia deliciosa) terhadap Kerusakan Histologis Sel Hepar Mencit (Mus musculus) Akibat Pemberian Parasetamol

Tujuan Penelitian : Penelitian ini bertujuan untuk mengetahui pengaruh sari buah kiwi (Actinidia deliciosa) dalam mengurangi kerusakan histologis sel hepar mencit (Mus musculus) akibat pemberian parasetamol. Metode Penelitian : Penelitian ini bersifat eksperimental laboratorik dengan post test only controlled group design. Sampel berupa mencit (Mus musculus) jantan, galur Swiss webster berumur 2-3 bulan dengan berat badan ± 20 gram. Sampel sebanyak 30 ekor dibagi dalam 3 kelompok, masing-masing kelompok terdiri dari 10 ekor mencit. Teknik sampling yang dipakai adalah incidental sampling. Kelompok kontrol (K), mencit diberi aquades 0,5 ml peroral perhari selama 14 hari. Kelompok perlakuan 1 (PI) mencit diberi aquades 0,5 ml peroral perhari selama 14 hari dan parasetamol dosis 5,07 mg/20gBB mencit pada hari ke 12,13 dan 14. Kelompok perlakuan 2 (PII), mencit diberi sari buah kiwi dosis 0,78g/20gBB mencit peroral setiap hari selama 14 hari dan parasetamol dosis 5,07 mg/20gBB mencit pada hari ke 12,13 dan 14. Hari ke-15 mencit dikorbankan kemudian hepar mencit dibuat preparat dengan metode blok parafin dan pengecatan Hematoksilin Eosin (HE). Gambaran kerusakan histologis sel hepar mencit diamati dan dinilai berdasarkan jumlah inti yang mengalami piknosis, karyoreksis, dan karyolisis yang dihitung dari 100 sel hepar di sentrolnuler. Data dianalisis dengan menggunakan uji One-Way ANOVA (α = 0,05) dan dilanjutkan dengan uji Post Hoc Multiple Comparisons (LSD) (α =0,05). Hasil Penelitian : hasil uji One-Way ANOVA menunjukkan adanya perbedaan yang bermakna antara ketiga kelompok perlakuan. Hasil uji LSD menunjukkan adanya perbedaan yang bermakna antara K-PI, PI-PII, dan K-PII. Sample consisted of male mice (Mus musculus) Swiss Webster, 2-3 months old with body weight ± 20 grams. Sample as many as 30 mice were divided into 3 groups, each group consisted of 10 mice. The sampling technique was incidental sampling. The control group (K), mice were given aquadest 0,5 ml peroral per day for 14 days. Treatment group 1 (PI) mice were given aquadest 0,5 ml peroral per day for 14 days and paracetamol dose 5.07 mg/20g weight of mice on day 12, 13 and 14. Treatment group 2 (PII), mice were given doses of kiwi essence 0.78 g/20g weight of mice per oral every day for 14 days and paracetamol dose 5.07 mg/20g weight of mice on day 12, 13 and 14. Finally on day 15th, mice were sacrificed and the liver of mice was made preparations with paraffin blocks methods and Hematoxylin eosin (HE) staining. Damage liver cells of mice were observed and assess based on number of pygnosis, karyorhexis and karyolytic nuclear cells from 100 liver cells in centrolobuler. Data were analyzed with One-Way ANOVA and continued with Post Hoc Multiple Comparisons test (LSD) (α = 0.05). Results: The result of One-Way ANOVA test showed significant difference between the three treatment groups. The result of LSD test showed a significant difference between the K-PI, P1-PII, and K-PII. Conclusions: According to this research, we concluded that the feeding of kiwi essence was able to decrease the liver cell damage of mice due to exposure of paracetamol.
