Efek hepatoprotektor ekstrak kubis (brassica oleracea) terhadap kerusakan sel hepar mencit yang diinduksi parasetamol

Tujuan Penelitian: Penelitian ini bertujuan untuk mengetahui efek pemberian ekstrak kubis dalam mencegah kerusakan sel hepar mencit yang diinduksi parasetamol dan efek peningkatan dosis ekstrak kubis dalam meningkatkan daya proteksi terhadap kerusakan sel hepar mencit yang diinduksi parasetamol.
Metode Penelitian: Penelitian ini bersifat eksperimental laboratorik dengan the post test only controlled group design. Sampel 28 ekor mencit jantan, galur Swiss webster, umur 2-3 bulan, berat badan + 20 gram. Teknik pengambilan sampel simple random sampling. Sampel dibagi dalam 4 kelompok, masing-masing kelompok terdiri dari 7 ekor. Kelompok KN dan KP, mencit diberi aquades selama 14 hari. Kelompok P1, mencit diberi ekstrak kubis dosis I dan kelompok P2, mencit diberi ekstrak kubis dosis II selama 14 hari. Parasetamol diberikan pada kelompok KP, P1, dan P2 pada hari ke-12, 13, dan 14. Hari ke-15, mencit dikorbankan dan hepar mencit dibuat preparat dengan metode blok parafin dan pengecatan HE. Gambaran histologis hepar dinilai berdasarkan penjumlahan piknosis, karioreksis, dan kariolisis. Data dianalisis dengan menggunakan uji One-way ANOVA (α = 0,05) dan dilanjutkan dengan uji LSD (α = 0,05).
Hasil Penelitian: Mencit kelompok P2 menunjukkan nilai rerata kerusakan paling kecil, diikuti kelompok KN, P1 dan KP. Hasil uji One-way ANOVA menunjukkan bahwa terdapat perbedaan bermakna pada keempat kelompok. Hasil uji LSD menunjukkan adanya perbedaan bermakna pada semua pasangan kelompok mencit.
Simpulan Penelitian: Ekstrak kubis dapat mencegah kerusakan sel hepar mencit yang diinduksi parasetamol dan peningkatan dosis ekstrak kubis dapat meningkatkan daya proteksi terhadap kerusakan sel hepar mencit yang diinduksi parasetamol.
Objective: The objectives of this research are to know the effect of cabbage extract in preventing the liver cells damage of mice which is induced by paracetamol and the effect of increasing dose of cabbage extract to increase its protection effect to the liver cells damage of mice which is induced by paracetamol.
Methods: This was laboratory experimental research with the post test only controlled group design. Samples were twenty eight male mice, Swiss webster type, 2-3 months old, and + 20 grams of each weight. Samples were divided into 4 groups, each group has 7 mice. Sampling technique was simple random sampling. The group of KN and KP, mice were given aquadest for 14 days. The group of P1, mice were given a first dose of cabbage extract and the group of P2, mice were given a second dose of cabbage extract for 14 days. Paracetamol was given to groups of KP, P1, and P2, on the 12th, 13th, and 14th day. The 15th day, mice were sacrificed and livers were made preparations by the method of paraffin block and stained by HE. Liver histological features were assessed based on quantifying of pyknosis, karyorhexis, and karyolysis. Data were analyzed using the One-way ANOVA test (α = 0.05) and continued with LSD test (α = 0.05).
Results: The mice in group of P2 showed the lowest score of damage rate, followed by KN, P1 and KP. The One-way ANOVA test results showed that there was significant difference between the four groups. The LSD test results showed that there were significant differences among all paired groups.
Conclusion: Cabbage extract can prevent the liver cell damage of mice which is induced by paracetamol and the increasing dose of cabbage extract can increase its protection effect to the liver cell damage of mice which is induced by paracetamol.
