Efek Pemberian Ekstrak Etanol Bunga Brokoli (Brassica Oleracea L. Var. Botrytis L) terhadap Struktur Histologis Ginjal Mencit (Mus Musculus) yang Diinduksi Parasetamol

ABSTRAK
Latar Belakang: Ekstrak etanol bunga brokoli diketahui mengandung senyawa saponin, flavonoid, alkaloid, terpenoid, steroid, Cyano Hydroxy Butene (CHB), sulforafan dan iberin sebagai senyawa antioksidan. Penelitian ini bertujuan untuk membuktikan efek pemberian ekstrak etanol bunga brokoli terhadap kerusakan struktur histologis ginjal mencit yang diinduksi parasetamol dan untuk mengetahui adakah perbedaan efek renoprotektor dari dosis ekstrak yang berbeda.
Metode Penelitian: Penelitian ini bersifat eksperimental laboratorik dengan rancangan  post test only controlled group design. Sampel 30 ekor mencit dibagi dalam 5 kelompok, masing-masing kelompok terdiri dari 6 ekor. Teknik sampling yang dipakai adalah consecutive sampling. Mencit Kelompok Kontrol Normal (KKN) dan Kontrol parasetamol (KKparaset) diberi akuades selama 14 hari. Mencit Kelompok Perlakuan 1 (KP1), Perlakuan 2 (KP2) dan Perlakuan 3 (KP3) diberi ekstrak etanol bunga brokoli dengan dosis masing-masing 42 mg/20 gBB; 56 mg/20 gBB dan 70 mg/20 gBB. Parasetamol diberikan pada kelompok KKparaset, KP1, KP2 dan KP3 pada hari ke 12, 13, dan 14. Pada hari ke-15, mencit dikorbankan dan ginjal mencit dibuat preparat dengan metode Blok Parafin dan pengecatan Hematoksilin Eosin (HE). Gambaran struktur histologis ginjal mencit dinilai berdasarkan penjumlahan inti sel epitel tubulus proksimal yang mengalami piknosis, karioreksis dan kariolisis. Data dianalisis menggunakan uji One-Way ANOVA dan dilanjutkan uji Post Hoc Multiple Comparisons LSD (α = 0,05).
Hasil Penelitian: Rerata kerusakan sel epitel tubulus proksimal ginjal pada KKN, KKparaset, KP1, KP2 dan KP3 berturut-turut adalah 6,08 ± 1,505 ; 36,00 ± 2,763. 30,42 ± 2,065; 23,67 ± 1,826; dan 14,92 ± 1,621. Hasil uji One-Way ANOVA dilanjutkan Post Hoc Multiple Comparisons LSD menunjukkan adanya perbedaan yang bermakna antara KKN-KKparaset, KKparaset-KP1, KKparaset-KP2, KKparaset-KP3, KP1-KP2, KP2-KP3, KP1- KP3, KKN-KP1, KKN-KP2 dan KKN-KP3.
Simpulan : Ekstrak etanol bunga brokoli memiliki efek proteksi terhadap struktur histologis ginjal mencit yang diinduksi parasetamol. Peningkatan dosis ekstrak etanol bunga brokoli dapat meningkatkan efek renoprotektor.

ABSTRACT
Background: The Ethanolic extract of broccoli flowers contains saponin, flavonoid, alkaloid, terpenoid, steroid, cyano hydroxy butene, sulforafan and iberin as an antioxidant compounds.  The objectives of this research are to know the effect of ethanolic extract of broccoli in preventing the renal histologist structure damage in mice was induced by paracetamol. And also to know is there any difference renoprotector effect in difference doses.
Methods: This was laboratory experimental with the post test only controlled group design. Samples were 30 mices devided into 5 groups, each group has 6 mices. Sampling technique in this research was consecutive sampling. Mices of normal control group KKN and parasetamol control group (KKparaset) will be given aquadest for 14 days. The first (KP1), second (KP2) and third (KP3) treatment group will be given with ethanolic extract of broccoli flowers dose 42 mg/20 gBB, 56 mg/20 gBB and 70 mg/20 gBB for 14 days. While the paracetamol in toxic dose was given on to KKparaset, KP1, KP2 and KP3 on day 12, 13, and 14.  On day 15, the mices was terminated  and the kidneys was made preparations by paraffin block methods and HE staining. Kidneys histological structure features assessed by sum of proximal tubule cells of renal damage that undergo pycnosis, karyorexys and karyolysis. Data were analized by One-Way ANOVA test (α = 0,05) and continued by Post Hoc Multiple Comparisons LSD test (α = 0,05).
Results: Mean of proximal tubulus epitel cells of renal damage on KKN, KKparaset, KP1, KP2 and KP3 was 6.08 ± 1.505 ; 36.00 ± 2.763. 30.42 ± 2.065; 23.67 ± 1.826; dan 14.92 ± 1.621. The result of One-Way ANOVA and Post Hoc Multiple Comparisons LSD showed there was significant difference between group of KKN-KKparaset, KKparaset-KP1, KKparaset-KP2, KKparaset-KP3, KP1-KP2, KP2-KP3, KP1- KP3, KKN-KP1, KKN-KP2 dan KKN-KP3.
Conclusion: The ethanolic extract of broccoli flowers showed renoprotector effect against paracetamol-induced renal histologic structure in mice. The increasing dose of ethanolic extract of broccoli can increase renoprotector effect.
