Dendrobium stratiotes is an orchid that has the potential to be developed because it has economic value with a variety of shapes, colors, sizes and distinctive aromas as well as long-lasting flower quality. Orchid propagation through in vitro culture supported by the application of appropriate plant growth regulators can overcome the problem of the length of time needed for orchid seeds to germinate. This study aimed to determine the exact interaction and concentration between kinetin and mung bean sprout extract on the growth of Dendrobium stratiotes orchids in vitro. The research was located at the Laboratory of Plant Physiology and Biotechnology, Faculty of Agriculture, Universitas Sebelas Maret from July to December 2022. The method used was a Completely Randomized Design (CRD) which was arranged in a factorial and consisted of 2 factors. The first factor was the kinetin concentration (0, 0,5, 1, 1,5, and 2 ppm). The second factor was mung bean sprout extract (0, 50, 100, 150, and 200 g.L -1 ). The two treatments then obtained 25 treatment combinations and each was repeated 5 times. The data obtained were analyzed using analysis of variance ? 5% if there was a significant difference then followed by the Duncan’s Multiple Range Test at 5% level and regression test. The result showed that the application of various combinations of kinetin and mung bean sprout extract could stimulate growth in height, number of leaves, number of roots, and fresh weight. The combination of 1 ppm kinetin + 200 g.L -1 mung bean sprout extract gave the best results in increasing the number of roots, while 1 ppm kinetin + 0 g.L -1 mung bean sprout extract could increase height, number of leaves, and fresh weight.
