ABSTRAK Hepar Mencit (Mus Musculus) Akibat Paparan Parasetamol. Skripsi, FakultasKedokteran, Universitas Sebelas Maret, Surakarta.Tujuan penelitian. Madu mengandung antioksidan yang dapat menangkal radikalbebas dan mengurangi terbentuknya NAPQI sebagai hasil dari metabolismeparasetamol. Penelitian ini bertujuan untuk mengetahui efek pemberian maduterhadap kerusakan sel hepar mencit akibat paparan parasetamol dan peningkatandosis madu dapat meningkatkan efek proteksinya terhadap kerusakan sel heparmencit akibat paparan parasetamol.Metode penelitian. Penelitian ini bersifat eksperimental laboratorik dengan posttest o nly controlled group design. Sampel berupa mencit (Mus musculus) jantan,galur Swiss webster berumur 2-3 bulan dengan berat badan + 20 gr. Sampelsebanyak 28 ekor dibagi dalam 4 kelompok, masing-masing kelompok terdiri dari7 ekor mencit. Teknik sampling yang dipakai adalah accidental sampling.Kelompok kontrol (K), mencit diberi aquades 0,2 ml peroral perhari selama 14hari. Kelompok perlakuan 1 (PI), mencit diberi aquades 0,2 ml peroral perhariselama 14 hari dan parasetamol dosis 0,1 ml/20 gr BB mencit pada hari ke-12, 13dan 14. Kelompok perlakuan 2 (PII), mencit diberi madu dosis 0,04 ml/20 gr BBmencit selama 14 hari dan parasetamol dosis 0,1 ml/20 gr BB mencit pada hari ke-12, 13 dan 14. Kelompok perlakuan 3 (PIII), mencit diberi madu dosis 0,08 ml/20gr BB mencit dan parasetamol dosis 0,1 ml/20 gr BB mencit pada hari ke-12, 13dan 14. Hari ke-15, mencit dikorbankan dengan cara neck dislocation kemudianorgan hepar diambil dan dibuat preparat dengan metode blok parafin danpengecatan Hematoksilin Eosin (HE). Gambaran histologis hepar diamati dandinilai berdasarkan kerusakan histologis yang berupa inti pyknosis, karyorrhexis,dan karyolysis, dimana dari setiap jenis kerusakan ini, masing-masing diberi skor1. Data dianalisis dengan menggunakan uji One-Way ANOVA (α = 0,05) dandilanjutkan dengan uji Post Hoc Multiple Comparisons (LSD) (α = 0,05).Hasil penelitian. Hasil uji Oneway ANOVA menunjukkan adanya perbedaan yangbermakna antara keempat kelompok perlakuan. Hasil uji Post Hoc menunjukkanadanya perbedaan yang bermakna antara P I-K, P I-P II, P II-K, P II-P III, PIII-Kdan menunjukkan perbedaan yang tidak bermakna antara kelompok P I-P III.Simpulan penelitian. Madu dapat mengurangi kerusakan sel hepar mencit (Musmusculus) akibat paparan parasetamol tetapi pada peningkatan dosis madu yangmelebihi dosis tertentu, tidak meningkatkan efek proteksinya terhadap kerusakansel hepar mencit.Kata kunci: madu, parasetamol, kerusakan sel hepar ABSTRACTHasan As’ari, G.0006090, 2009. The Influence of Honey to Liver Cell Damageof Mice (Mus musculus) as a Result of Induce Paracetamol. Script, Faculty ofMedicine, Sebelas Maret University, Surakarta.Objective: Honey has antioxidant as a protection of free radicals and reducingNAPQI which produced by paracetamol metabolism. The objective are to knowthe influence of honey to the liver cell damage of mice which is induced byparacetamol and the increase of honey dose can also increase protection effect tothe liver cell damage of mice which is induced by paracetamol.Methods: This was a laboratory experimental research with post test o nlycontrolled group design. Samples in this research were twenty eight male mice(Mus musculus), Swiss webster type, 2-3 months old and + 20 gr of each weight.Samples divided into 4 groups, each group has seven mices. Mice for controlgroup (K) will not be given paracetamol and honey, it was o nly given aquadest0,2 ml/20 gr weight of mice for 14 days in a row. The first treatment group (PI)will be given paracetamol with dose 0,1 ml/20 gr weight of mice o n the day 12, 13and 14. The second treatment group (PII) will be given honey dose I whichconsist of 0,04 ml/20 gr weight of mice for 14 days in a row and also paracetamoldose 0,1 ml/ 20 gr weight of mice o n day 12, 13 and 14. The third treatment group(PIII) will be given honey dose II which consist of 0.08 ml/20 gr weight of micefor 14 days in a row and also paracetamol dose 0,1 ml/ 20 gr weight of mice o nday 12, 13 and 14. Finally o n day 15th, mice were sacrificed with neck dislocation.After that, we made preparation from the liver that stained with HematoxillinEosin (HE). Preparation was observed and scoring based o n the liver histologicaldamage (karyopyknosis, karyorrhexis and karyolysis). And each o ne ofkaryopyknosis, karyorrhexis and karyolysis was given score 1. Data were analizedby One-Way ANOVA test (α = 0,05), and continued by Post Hoc MultipleComparisons (LSD) test (α = 0,05).Results: Result of One-Way ANOVA showed that there was a significantdifference between 4 groups. Result of LSD method showed that there was asignificant difference between K-PI, K-PII, K-PIII, PI-PII and PII-PIII groups, butthere was not the significant difference between group PI-PIII.Conclusion: According to this research, we concluded that the feeding of honeywas able to decrease the liver cell damage of mice (Mus musculus) but theincrease of honey dose which exceed certain dose was not followed by theincrease of protection effect to the liver cell damage of mice which was inducedby paracetamol.Key words : honey, paracetamol, liver cell damage.
