ABSTRAK

 

Hepar Mencit (Mus Musculus) Akibat Paparan Parasetamol. Skripsi, Fakultas

Kedokteran, Universitas Sebelas Maret, Surakarta.

Tujuan penelitian. Madu mengandung antioksidan yang dapat menangkal radikal

bebas dan mengurangi terbentuknya NAPQI sebagai hasil dari metabolisme

parasetamol. Penelitian ini bertujuan untuk mengetahui efek pemberian madu

terhadap kerusakan sel hepar mencit akibat paparan parasetamol dan peningkatan

dosis madu dapat meningkatkan efek proteksinya terhadap kerusakan sel hepar

mencit akibat paparan parasetamol.

Metode penelitian. Penelitian ini bersifat eksperimental laboratorik dengan post

test o nly controlled group design. Sampel berupa mencit (Mus musculus) jantan,

galur Swiss webster berumur 2-3 bulan dengan berat badan + 20 gr. Sampel

sebanyak 28 ekor dibagi dalam 4 kelompok, masing-masing kelompok terdiri dari

7 ekor mencit. Teknik sampling yang dipakai adalah accidental sampling.

Kelompok kontrol (K), mencit diberi aquades 0,2 ml peroral perhari selama 14

hari. Kelompok perlakuan 1 (PI), mencit diberi aquades 0,2 ml peroral perhari

selama 14 hari dan parasetamol dosis 0,1 ml/20 gr BB mencit pada hari ke-12, 13

dan 14. Kelompok perlakuan 2 (PII), mencit diberi madu dosis 0,04 ml/20 gr BB

mencit selama 14 hari dan parasetamol dosis 0,1 ml/20 gr BB mencit pada hari ke-

12, 13 dan 14. Kelompok perlakuan 3 (PIII), mencit diberi madu dosis 0,08 ml/20

gr BB mencit dan parasetamol dosis 0,1 ml/20 gr BB mencit pada hari ke-12, 13

dan 14. Hari ke-15, mencit dikorbankan dengan cara neck dislocation kemudian

organ hepar diambil dan dibuat preparat dengan metode blok parafin dan

pengecatan Hematoksilin Eosin (HE). Gambaran histologis hepar diamati dan

dinilai berdasarkan kerusakan histologis yang berupa inti pyknosis, karyorrhexis,

dan karyolysis, dimana dari setiap jenis kerusakan ini, masing-masing diberi skor

1. Data dianalisis dengan menggunakan uji One-Way ANOVA (α = 0,05) dan

dilanjutkan dengan uji Post Hoc Multiple Comparisons (LSD) (α = 0,05).

Hasil penelitian. Hasil uji Oneway ANOVA menunjukkan adanya perbedaan yang

bermakna antara keempat kelompok perlakuan. Hasil uji Post Hoc menunjukkan

adanya perbedaan yang bermakna antara P I-K, P I-P II, P II-K, P II-P III, PIII-K

dan menunjukkan perbedaan yang tidak bermakna antara kelompok P I-P III.

Simpulan penelitian. Madu dapat mengurangi kerusakan sel hepar mencit (Mus

musculus) akibat paparan parasetamol tetapi pada peningkatan dosis madu yang

melebihi dosis tertentu, tidak meningkatkan efek proteksinya terhadap kerusakan

sel hepar mencit.

ABSTRACT

Hasan As’ari, G.0006090, 2009. The Influence of Honey to Liver Cell Damage

of Mice (Mus musculus) as a Result of Induce Paracetamol. Script, Faculty of

Medicine, Sebelas Maret University, Surakarta.

Objective: Honey has antioxidant as a protection of free radicals and reducing

NAPQI which produced by paracetamol metabolism. The objective are to know

the influence of honey to the liver cell damage of mice which is induced by

paracetamol and the increase of honey dose can also increase protection effect to

the liver cell damage of mice which is induced by paracetamol.

Methods: This was a laboratory experimental research with post test o nly

controlled group design. Samples in this research were twenty eight male mice

(Mus musculus), Swiss webster type, 2-3 months old and + 20 gr of each weight.

Samples divided into 4 groups, each group has seven mices. Mice for control

group (K) will not be given paracetamol and honey, it was o nly given aquadest

0,2 ml/20 gr weight of mice for 14 days in a row. The first treatment group (PI)

will be given paracetamol with dose 0,1 ml/20 gr weight of mice o n the day 12, 13

and 14. The second treatment group (PII) will be given honey dose I which

consist of 0,04 ml/20 gr weight of mice for 14 days in a row and also paracetamol

dose 0,1 ml/ 20 gr weight of mice o n day 12, 13 and 14. The third treatment group

(PIII) will be given honey dose II which consist of 0.08 ml/20 gr weight of mice

for 14 days in a row and also paracetamol dose 0,1 ml/ 20 gr weight of mice o n

day 12, 13 and 14. Finally o n day 15th, mice were sacrificed with neck dislocation.

After that, we made preparation from the liver that stained with Hematoxillin

Eosin (HE). Preparation was observed and scoring based o n the liver histological

damage (karyopyknosis, karyorrhexis and karyolysis). And each o ne of

karyopyknosis, karyorrhexis and karyolysis was given score 1. Data were analized

by One-Way ANOVA test (α = 0,05), and continued by Post Hoc Multiple

Comparisons (LSD) test (α = 0,05).

Results: Result of One-Way ANOVA showed that there was a significant

difference between 4 groups. Result of LSD method showed that there was a

significant difference between K-PI, K-PII, K-PIII, PI-PII and PII-PIII groups, but

there was not the significant difference between group PI-PIII.

Conclusion: According to this research, we concluded that the feeding of honey

was able to decrease the liver cell damage of mice (Mus musculus) but the

increase of honey dose which exceed certain dose was not followed by the

increase of protection effect to the liver cell damage of mice which was induced

by paracetamol.
