Pengaruh paparan bising intermittent kronik terhadap CD8+ pada tikus putih (Rattus norvegicus)

× ABSTRAK Tujuan Penelitian : Bising merupakan stressor yang dapat berdampak negatif bagi tubuh. Apabila bising datang secara intermittent maka akan lebih memberikan pengaruh bagi tubuh dan dampaknya akan bertambah buruk jika bersifat kronik. Salah satunya terhadap psikoneuroimunologi manusia, yaitu dengan perubahan kadar kortisol darah didapatkan gangguan pada sistem imun seluler tubuh manusia. Penelitian ini bertujuan untuk mengetahui apakah ada pengaruh paparan kronik bising intermittent terhadap CD8+ pada tikus putih. Metode Penelitian : Penelitian ini bersifat eksperimen laboratorium dengan rancangan penelitian ‘the post test only control group’ design. Lokasi penelitian di rumah kaca, Laboratorium Sentral MIPA Biologi, Universitas Sebelas Maret, Surakarta. 30 ekor tikus putih (Rattus norvegicus) dipilih secara random dengan kriteria inklusi jantan, galur wistar, berat badan ± 150-250 gram, dan berumur 2-3 bulan, dibagi dalam 3 kelompok yaitu : Kelompok I (kontrol), Kelompok II (paparan dengan intensitas 72-75 dB), dan Kelompok III (paparan dengan intensitas 88-90 dB). Tikus putih diadaptasikan selama 7 hari kemudian kelompok II dan III diberi paparan bising secara intermittent (frekuensi 350 Hz) dengan pengaturan1 jam paparan, 1 jam istirahat selama 5 jam (11.00-16.00) dalam waktu 21 hari. Hari berikutnya, hewan uji tersebut diambil darah nya secara intrakardiak sebanyak 1cc (tiap kelompok hanya 9 ekor). Sampel darah kemudian ditambah dengan FITC anti-rat CD8+ antibody sebelum dianalisis dengan menggunakan flowcytometry di Laboratorium Patologi Klinik, Universitas Gadjah Mada. Data yang diperoleh diuji dengan uji statistik Oneway Anova dan dilanjutkan dengan uji LSD. Hasil Penelitian : Hasil uji Oneway Anova menunjukkan adanya perbedaan yang bermakna di antara ketiga kelompok yaitu p=0,000 (p<0,05). Pada uji LSD juga diadapatkan perbedaan bermakna antara ketiga kelompok Simpulan : Paparan kronik bising intermittent mempunyai pengaruh terhadap kadar CD8+ pada tikus putih (Rattus norvegicus). ABSTRACT Objective : Noise is a stressor which cause physiological, psychological and behavioral changes in human. Intermittent noise has negative effect on the body and overstimulation due to chronic exposure may affect ones health. One of its effect is on human psychoneuroimmunology that can be seen by the changes of cortisol level which can cause a disturbance to cellular immune system. The purpose of this study was to determine the effect of chronic intermittent noise exposure on CD8+ count in rat. Methods : This was an experimental research using posttest only control group design. This study took place in the green house of MIPA Central Laboratory (Biology), Sebelas Maret University. Thirty male rats (Rattus noevegicus) was randomly selected, aged 2-3 months old and weighed 150-250 grams were divided into 3 groups, Group I (control), Group II (exposed to 72-75 dB of sound intensity) and Group III (exposed to 88-90 dB of sound intensity). They were adapted to the laboratory environment for 7 days before the start of experiment. After that, group II and III were exposed intermittently to the 350 Hz noise with an hour exposure and an hour break for 5 hours per day within 21 days. On the 22nd day, 1cc blood was taken intracardial (only 9 samples per group). Then the blood samples were added with FITC anti-rat CD8+ antibody before they were analysed by flowcytometry in Laboratory of Clinical Pathology, Gadjah Mada University. Data were analyzed using Oneway Anova Test and LSD test. Results : Result from Oneway Anova test showed that there was a significant difference among the three groups at p=0,000 (p<0,05). LSD test showed that there were significant differences among groups. Conclusion : Chronic intermittent noise exposure can influence the CD8+ count in rat (Rattus norvegicus).
