ABSTRAKLatar Belakang: Kanker merupakan kelainan genetik yang melibatkan perubahan dinamikgenom sehingga menyebabkan pertumbuhan sel tidak terkendali, mampu menginvasi danbermetastasis. Kanker payudara merupakan kanker yang paling banyak ditemukan pada wanita.Banyak sekali kandungan senyawa bioaktif fitokimia yang ditemukan dalam tanaman sirsak.Salah satu komponen bermanfaat yang terpenting adalah annonaceous acetogenins. Penelitianini untuk menguji efek sitotoksik ekstrak ethanol daun sirsak pada cell line Ca Mammae (T47D).Metode Penelitian: Jenis penelitian ini adalah eksperimental laboratorik dengan post test controlgroup deisgn. Sampel pada penelitian ini adalah cell line Ca Mammae (T47D) yang dibagi dalamtiga kelompok, yaitu kelompok perlakuan ekstrak ethanol daun sirsak, kelompok pemberiandoksorubisin dan kelompok kontrol sel. Efek sitotoksik diukur dengan MTT assay kemudiandianalisis untuk menentukan Inhibitory concentration fifty (IC50).Hasil Penelitian: IC50 ekstrak ethanol daun sirsak pada cell line Ca Mammae (T47D) adalah69,250 ?g/ml dan pada pemberian doksorubisin adalah < 0,195 ?g/ml.Simpulan Penelitian: Berdasarkan hasil penelitian, ekstrak ethanol daun sirsak memiliki efeksitotoksik yang lemah (weak) pada cell line Ca Mammae (T47D) dengan IC50 69,250 ?g/ml.Keywords: MTT assay, Ekstrak ethanol daun sirsak, sitotoksik, Cell Line Ca Mammae (T47D)ABSTRACTBackground: Cancer is a genetical disorder which involves dynamic changes of genome, thus itcauses abnormal cell growth, invasion and metastasis of cell.
