Abstrak

Purpose: The aim of this study is to evaluate both functionally and histologically the validity of the Amniotic Membran for nerve regeneration in rat sciatic nerve crush injury.

Materials & methods: In this study 18 healthy Sprague Dawley rats were used. In all rats, a unilateral right side sciatic nerve crush injury was performed by pinched by forcep and observed by different methods: Group I (control group): included 9 rats, the damaged nerve was left without amniotic membrane. Group II: included 9 rats, , the damaged nerve was left with amnion membrane. both functionally and histologically evaluation was performed over 21 days postoperatively.

Results: Functional results showed that there was significant difference of walking track analysis between group I and group II on day 5,7,14, and 21 (P<0.05). Histological results showed that after 21 days there was  complete nerve regeneration and reconnection in the Group II. Histological results showed that there was significant difference between group I and group II for Axonal degeneration (P:0.0024), Nerve regeneration (P:0.016), and lower inflamation reaction (P:0.016). But there was not significant difference in Nerve diameter (P:0.23).

Conclusion: We can conclude that application of amniotic membrane in nerve crush injury damage nerve was better than damaged nerve that left without amniotic membrane both functionally and histologically and could be used as an alternative technique to improve nerve healing in nerve crush injury
