Matoa has become a defining feature of Papuan flora. Matoa fruit is well-known because it tastes like a cross between longan, rambutan, and durian. The matoa plant was used by the community for a number of purposes, including the use of the stems in the wood industry, the consumption of the fruit, and the use of the leaves and seeds in medicine. Matoa has tremendous potential and benefits but it has not been widely cultivated. This is due to seed storage and plant propagation limitations. Attempts to solve this problem are being made through tissue culture matoa propagation. Tissue culture is used to produce large quantities of the same, faster, and high-quality seedlings. The goal of this research is to develop vegetative planting material that can be propagated in vitro.

The research took place at the Laboratory of Plant Physiology and Biotechnology Culture, Faculty of Agriculture, Sebelas Maret University, from June 2021 to January 2022. This study employed a completely randomized design (CRD) with a factorial arrangement of 2x4 treatment factors, namely types of concentrations of ZPT, IAA, and BAP, resulting in 16 treatment combinations and three trays per combination. The first factor is IAA concentration, which has four levels: 0 ppm, 0.5 ppm, 1 ppm, and 1.5 ppm. The second factor is BAP concentration, which has four levels: 0 ppm, 2 ppm, 4 ppm, and 6 ppm. MS media (Murashige and Skoog) used in this research, which modified with the addition of growth regulators IAA and BAP. The research implemented using sterilization of equipment, manufacture of treatment media, sterilization of explants, planting of explants, observation, and maintenance. The obtained data were analyzed using the F test at the 5% level. If there is a significant difference, the DMRT (Duncan Multiple Range Test) completed at the 5% level.

The results of the research showed that the time of emergence of roots, time of emergence of leaves, number of roots, and number of leaves had a high growth rate. The existence of a relationship between the number of leaves and stem nodes indicates that the growth of matoa explants is interrelated.
